Lanprol

📌 COMPOSITION

Each capsule contains: Lansoprazole USP 15, 30 mg (as enteric coated pellets).

📌 DESCRIPTION

Lanprol (Lansoprazole) suppresses gastric acid secretion by specific inhibition of the (H+/K+ - ATPase) enzyme system at the secretory surface of the gastric parietal cells. Lansoprazole is rapidly absorbed after oral administration and peak plasma concentrations are achieved about 1.5 hours. Bioavailability is 80% or more even with the first dose. It is extensively metabolised in the liver and excreted in urine. The plasma half‑life is around 1.4 hours, but the duration of action is much longer. Lansoprazole is about 97% bound to plasma proteins.

📌 INDICATIONS

Lanprol is indicated in the treatment of:

  • Duodenal ulcer, Gastric ulcer and Acid‑related dyspepsia.
  • Gastro‑oesophageal reflux and Zollinger‑Ellison syndrome.

📌 DOSAGE & ADMINISTRATION

Lanprol is preferred to be taken once daily in the morning before food.

  • Duodenal ulcer: 30 mg for 4 weeks. Maintenance dose 15 mg daily.
  • Gastric ulcer: 30 mg daily for 8 weeks.
  • Acid‑related dyspepsia: 15-30 mg for 2-4 weeks.
  • Gastro‑oesophageal reflux: 30 mg for up to 8 weeks. Maintenance dose 15-30 mg.
  • Zollinger‑Ellison syndrome: 60 mg once a day. Daily dosage of more than 120 mg should be administered in divided doses.

📌 RESTRICTIONS ON USE

Contraindications

Hypersensitivity to Lansoprazole.

Precautions
  • Symptomatic response to therapy with Lansoprazole does not preclude the presence of gastric malignancy.
  • Consider dose reduction in severe hepatic impairment.
Use in pregnancy and lactation

There are no adequate and well‑controlled studies on its use.

📌 DRUG INTERACTIONS

  • Lansoprazole may interfere with the absorption of drugs whose bioavailability is dependent on gastric pH such as ketoconazole, ampicillin, iron salts and digoxin.
  • Lansoprazole increases the clearance of theophylline.
  • Sucralfate delays the absorption and bioavailability of Lansoprazole.

📌 ADVERSE EFFECTS

Lansoprazole is well tolerated in both short‑term and long‑term trials. Reported adverse effects include diarrhoea, abdominal pain, nausea and headache.

📌 STORAGE INSTRUCTIONS

  • Store below 30°C, protected from light and humidity.
  • Close the bottle firmly after each use.
